Chuck Noland (Tom Hanks) is een hooggeplaatste functionaris bij FedEx die zijn werk boven zijn privéleven stelt. Terwijl hij met een vrachtvliegtuig meevliegt komt het vliegtuig in een storm terecht. Als dan een deel van de lading ontploft, stort het vliegtuig in de Stille Oceaan neer. Chuck weet te ontsnappen uit het vliegtuig terwijl het zinkt en spoelt aan op een eiland. Terwijl zijn naasten in de veronderstelling leven dat hij dood is, bedenkt hij manieren om te overleven en te ontsnappen.
